چکیده مقاله:

Background γ-Aminobutyric acid (GABA) is considered to be the predominant inhibitory neurotransmitter in mammalian central nervous systems (CNS). There are two major classes of GABA receptors: GABAARs and GABABRs. The GABAA receptor is derived from various subunits such as alpha1-alpha 6, beta1-beta 3, gamma1-gamma 4, delta, epsilon, pi, and rho1-3. Intensive research has been performed to understand and establish the distribution and functions of these receptors in the CNS and peripheral tissues. The presence of some GABAA receptors in sperm prompted us to explore the existence of GABAA receptors in rat testis and sperm. MaterialsAndMethods Total cellular RNA was isolated from Wistar rat sperm and testis and reverse transcriptased to cDNA. PCR reactions were performed in a 20μl (microliter) volume containing specific GABAAR subunits (forward and reverse primers) with other required materials. Reactions were carried out using a PCR machine to investigate the existence of GABAA receptor subunits in rat testis and sperm. The amplification products were analyzed on 2% agarose gels stained with ethidium bromide. Results Our results showed that GABAARs composed of α5, β1, β3, and γ1 subunits were expressed in both testis and sperm. These results indicate that, in sperm, GABAA receptors might have important functions. Conclusion Sperm could be a novel site of GABAA expression. Further studies should be taken to explore the role of these receptors on sperm acrosome reaction.

Quantitative Analysis of GABAA Gamma Receptor Subunits in the Developing Embryonic Chick Forebrain

Objective(s) In this study we investigated the expression of GABAA receptor subunits during brain development. These receptors may change in the embryonic chick forebrain. Materials and Methodes The expression levels of four types of GABAA receptor gamma subunits (γ1, γ2, γ3 and γ4) were quantified in the embryonic chick forebrain at 32 hr, 3, 7, 14, and 20 days of incubation and day one aft...

Hippocampal GABAA Receptor and Pain Sensitivity during Estrous Cycle in the Rat

Background: Estradiol and progesterone as well as hippocampal GABAA receptors are believed to play a role in the modulation of pain. The aim of present study was to investigate the effect of intrahippocampal injections of GABAA receptor agonist (muscimol) and GABAA receptor antagonist (picrotoxin) on pain sensitivity during estrous cycle. Methods: Pain sensitivity was evaluated in rats ...

Age-related decrease of GABAA receptor subunits and glutamic acid decarboxylase in the rat inferior colliculus.

A selective age-related decrease in both the protein and mRNA levels of the most abundant GABAA receptor subunits has been revealed in the rat inferior colliculus. The number (not affinity) of the native and fully assembled GABAA receptors assayed by 3H-muscimol binding was also decreased (35-49%). The decrease in GABA receptors was accompanied by a decrease in the protein and mRNA of the GABA-...

Function and pharmacology of multiple GABAA receptor subunits.

LYl7l555. SinLe forskolin activates adenylyl cyclase, effects of Dr receptors on Na'/X+-ATPase would then be mediated via protein phosphorylation and CAMP-dependent protein kinase. There are reports that Na+/K+-ATPase may be regulated by phosphoryl-ation, and CAMP has been reported to inhibit the enzyme via a cAMP-&pendent protein kinase7, consistent with the scheme outlined above. However, it ...

GABAA receptor subunits identified in Paramecium by immunofluorescence confocal microscopy.

The presence of opioid, beta-adrenergic and cholinergic receptors has been demonstrated in ciliated protozoa, but little is known about gamma-aminobutyric acid (GABA) receptors. In this study we have analyzed the distribution of GABA(A)-type receptor subunits in Paramecium. Confocal laser microscopy using antibodies specific for alpha(1)-, alpha(2)-, alpha(3)-, alpha(6)-, beta(2/3)-, gamma(2)-,...
